Police believe a semi-trailer jack-knifed on the Melba Highway which caused it to collide with two other vehicles heading in the opposite direction around 8.20pm on Sunday night.
A Dandenong woman, aged in her 50s, who was the sole occupant of a Mitsubishi van, died at the scene.
A 29-year-old woman from Pakenham Upper was driving a Nissan Navara and towing a horse float.
